It’s a fairly common surname in Co. Tyrone. Looking at the 1901 census of the county, there were 145 of them then, scattered across the county.
To have any real chance of tracing your ancestors you need to know their exact denomination and have some idea of where in the county they lived.
Looking at the 1901 census, the vast majority were Presbyterian, with some Church of Ireland (and related denominations) plus about 1 RC family. So the location, timing of their departure to the USA (ie early 1800s when it was mainly Protestants who were leaving Ireland), their Scottish surname and religious denominations suggest that most were of Scottish origins and would have arrived in Ireland in the 1600s as part of the Plantation.
Very few churches in Tyrone have any records for the late 1700s/early 1800s and so the chances of finding any documentary record of this family are not high.

I have looked at the Parish Church records for Saint Luran's Parish, Derryloran, Co Tyrone
(http://freepages.rootsweb.com/~cotyroneireland/genealogy/churchrecord/saintluran.html)
and have found that in the record for 1797 a Robert Glasgow was shown as the Church Warden.
The records only start at 1797 so you can't go any further back. I would advise you to be aware that the person holding the pen is the one who decides on spelling. I have seen the surname Glasco.
There is a Baptism of James, son of William and Mary Glasgow on 21 Aug 1799.
The name Glasgow is also mentioned in the indexes 1810-1896.
Elwyn has mentioned the 1901 Census for Tyrone (http://www.census.nationalarchives.ie/search/) and I noticed that Parish of Derryloran is in the DED (District Electoral Division) of Cookstown. Unfortunately, the oldest people are only in their late 70s, but naming patterns may be of some help to you.
I have also looked at the Griffith's Valuation for Tyrone. (http://www.askaboutireland.ie/griffith-valuation/index.xml?action=nameSearch)
There are 61 entries printed in 1859, some 40 years before the 1901 Census. These are for people who were either tenants or property owners of land (over 1 acre, I think). When you click on Details you will get info on the owner, lessor, full location data etc. You can also bring up maps showing location.
If you go to Griffith's Places, you will be given the Placenames of the parish, with their full location.
If you then click on the Occupants button you will be shown the names of the people who lived in that Place name. (neighbours).

Have you actually found your relative landing in America or any other information as to where he settled, died etc?

Thank u so much. I have found the same info u have presented here. I am trying to find Robert and Elizabeth marriage and didnt see any in the church records of Derryloran where the majority of Glasgow lived. I could not find any mention of a parish or county in Usa and have done a exhaustive search so at this point its all speculation. I know everything about Robert and Elizabeth in the states and their descendants. This is my 5x grandparents but i do not know Elizabeth surname which is the tree branch i am totally missing. With the naming pattern, John would be Robert father and William Elizabeth father. Margaret would be robert mother and Nancy is Elizabeth mother. I uave done dna but no Glasgow surnames match
